DETERMINATION OF DISULFIRAM-CONTAINING PHARMACEUTICALS BY
IR SPECTROSCOPY AND HİGH PRESSURE LIQUID CHOROMATOGRAPHY

Summary:
In this study, infrared (IR) spectroscopic and high pressure
liquid chromatographic (HPLC) methods are described for
quantitative determination of disulfiram (DSF) in solid
dosa¬ge form. IR spectroscopic method (KBr disc technique)
was used and dehydrocholic acid (DHCA) as inteenal standard.
The spe¬cific absorption bands at 914 and 1705 cm-1 were
chosen for DSF und DHCA respectively. Beer’s Law was obeyed
in the con¬centration range of 0.4-1.2% w/w in KBr. At
914 cm-1, regres¬sion equation was found to be y=0.2267x
+ 0.1287 (r=0.9976)
In HPLC method, DSF and Me.frusid (MFD) (internal standard)
were separated by isocratic system in that mobile phase
consis¬ted of methanol-phosphate buffer (pH=3±0.1) (80:20
v/v). Fluw-rate was 0.7 ml. min-l and Luna 5?C18 (250
x 4.6 mm) was used as stationary phase. Linear concentration
was obta¬ined as 1-4mcg mL-1. The regression equation
was found to be y= 5.1504x - 0.272 (r= 0.9989)
