OF GEONETICS ON THE BIOAVAILABILITY AND DISPOSITION KINETICS
OF MEFENAMIC ASID

Summary:
The effect of geonetics on the pharmacokinetic behavior
of mefenamic acid after an oral administration of drug
was studied in 27 healty rabbits. Mefeamic acid was estimated
by high performace liquid chromatography. The Cmax of
3.36 ? 0.55 ?g/mL was attained within 2.5 hours. The values
for AUC0-? and MRT of the drug were noted to be 14.9 ?
2.02 ?g.hr/mL and 4.16 ? 0.26 hours, respectively. Elimination
half-life, volume of distribution and total body clearance
were observed to be 1.93 ? 0.29 hrs, 14.5 ? 2.58 L and
86.6 ? 9.67 mL/min., respectively. Remarkable variations
were noted when the present findings were compared with
the respective values cited in the literature. Inadequate
studies on the pharmacokinetics of mefenamic acid made
it difficult to conclude that geonetics is the only basis
of these variations. The other variation factors may include
differences in study design, subject selection, drug dose,
dosage forms, routes of administration, analytical methods
and species difference. The use of various pharmacokinetic
computational techniques and pharmacokinetic software
may also be the variation determinant.
